HPLC-MS Determination of 5 Polyphenols in Shells of Camellia Oleifera

Abstract
HPLC-MS was applied to the determination of 5 polyphenols (i.e. gallic acid,catechin,epicatechin,rutin and quercetin) in shells of camellia oleifera. The sample was extracted with methanol. Thermo Scientific Syncronics C18 chromatographic column was used as stationary phase,and mixtures of acetonitrile and formic acid (1+99) solution mixed in different ratios were used as mobile phase in gradient elution. ESI- and multi-reaction monitoring were adopted in MS. Linear relationships between values of peak area and mass concentration of the 5 polyphenols were kept in definite ranges. Detection limits found were ranged from 0.02 to 0.50 mg·L-1. The proposed method was applied to the analysis of sample of camellia oleifera shell. Values of recovery found by standard addition method and RSD′s (n=5) found were in the ranges of 90.0%-105% and 1.1%-3.4%,respectively.
